Biotype identification of Spodoptera frugiperda individuals from the Northeast region of Tamaulipas, Mexico

Biotypes in Spodoptera frugiperda individuals have been characterized as corn and rice biotypes, which may be associated to host plant. In this work a molecular identification of biotypes of S. frugiperda larvae was determined through the technique of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Samples were collected from maize fields in the city of Reynosa, located at the Northeast region of the state of Tamaulipas. Samples were transferred to the laboratory for DNA extraction using a commercial kit from Promega. The amplification of cytochrome oxidase I gene was performed with primers, forward JM76 (5'GAGCTGAATTAGG (G / A) ACTCCAGG3 '), and reverse JM77 (5'ATCACCTCC (A / T) CCTGCAGGATC3'). Digestion was conducted with the enzyme MspI. Results indicated amplification products of 569 base pairs. Digestion products corresponded to 497 and 72 base pairs, which are characteristic of corn biotype, which was found in all individuals. No rice biotype was found. Therefore, we could distinguish the presence of corn biotype of S. frugiperda in the city of Reynosa, Tamaulipas in maize crops.
