Application of RNAi to control of tobacco whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae) and target gene selection

 Tobacco whitefly, Bemisia tabaci, is major pest that feeds over 600 plants. The most general method to control of tobacco whitefly is chemical control using insecticides, but this method have resistance problem. So, in this study, we try to control tobacco whitefly using RNAi. First of all, to select target gene for using RNAi, we construct cDNA library of tobacco whitefly and conduct LR recombination to tobacco rattle virus (TRV) RNA2 vector added att site to induce RNAi. To select genes that have effect to control of tobacco whitefly, we inoculated random genes to plant, and induced feeding of tobacco whitefly. Consequently, three unknown genes were selected. So, we checked whether or not three genes expressed in plant and gene expression changed by RNAi in insect through qRT-PCR. It is suggested that final selected target gene may be applied to control of tobacco whitefly using RNAi.
