A new transcriptomics resource for Coleoptera – next generation sequencing of the pollen beetle, Meligethes aeneus Col. Nitidulidae

We present the first sequenced transcriptome of pollen beetle, Meligethes aeneus F. (Coleoptera: Nitidulidae), the major univoltine pest of oilseed rape in many European countries. By using Roche 454 pyrosequencing and Illumina sequencing of cDNA libraries from mixed-sex adults of 4 different pollen beetle populations we were able to assemble a reference transcriptome and to identify differentially expressed unigenes between pyrethroid susceptible and resistant populations. Genes of interest (GOI) in terms of insect toxicology, i.e. molecular target sites of insecticides and detoxification enzymes such as cytochrome P450s were identified and further analyzed. A phylogenetic analyses of GOI was performed by comparison with transcriptome/genome data available for other insects, particularly coleopteran species such as Tribolium castaneum and Dendroctonus ponderosae.
