Fishing for the endosymbionts of the glassy-winged sharpshooter, Homalodisca coagulata (Homoptera:Cicadellidae)

Fluorescent in situ hybridization (FISH) was used to examine the intracellular endosymbionts of the glassy-winged sharpshooter (GWSS), Homalodisca coagulata. The endosymbionts were contained within bacteriocytes, forming organs referred to as bacteriomes, which consisted of paired reddish organs associated with yellow glandular-like structures located in the ventro-lateral anterior portion of the abdomen. One ribosomal DNA probe was used in order to demonstrate the presence of all eubacteria and three specific probes were used to demonstrate a, b and g proteobacteria. The red portion of the bacteriome contained only spherical organisms, while sections of the yellow portion contained several morphological forms. Correlation between specific probes, morphology and species level identification will be discussed.
