Genome sequencing of the Cotesia flavipes polydnavirus

Diatraea saccharalis is a key pest of sugarcane, but a minor pest of sweet corn, rice, and shorgum. Natural enemies of D. saccharalis have been reported to be important in reducing its populations, thus minimizing damage to crops. Cotesia flavipes is a gregarious parasitoid of D. saccharalis. The parasitoid injects polydnavirus (PDV) genes into D. saccharalis, which are transcribed into proteins that alter the physiology and suppress the immune system of the host. This paper reports the establishment of a D. saccharalis-C. flavipes colony. Polydnavirus DNA from ovaries of C. flavipes females was isolated and purified using two extraction protocols. DNA sequencing of each segment of the C. flavipes PDV genome is under way at the Institute for Integrative Genome Biology in UCR. The final goal is to identify potential opening reading frames of immune-suppresive genes and others with significant physiological functions. This research was jointly funded by CONACYT-UC MEXUS at UC-Riverside and IPN in Reynosa, Mexico.
