A survey for internal intracellular and extracellular bacteria within larval and adult Culicine mosquitoes: Molecular identification using 16S rRNA

The purpose of surveying the normal microbiota of members within the family Culicidae is to gain a more complete understanding of mosquito-bacteria interactions. The parameters for our study included: life stage (larva and adult), representatives of Culicidae, adult gender, body segment, and environment. Identification of the bacteria was achieved by PCR of 16S rRNA, Denaturing Gradient Gel Electrophoresis (DGGE), and by sequencing of DNA. We found that late instar larvae contained a more diverse group of bacteria compared to that found in young adults (1-3 days). Older mosquitoes (4+ days), however, harbored a diverse group of bacteria internally. We also found that the bacterial community in males differed from that of females. This information can be applied to mosquito control strategies, such as Sterile Insect Technique (SIT) and paratrangenesis.
