Preliminary analysis of mtDNA sequence data for Anoplophora: Implications for identification of species boundaries and source population determinations of Asian longhorned beetle (A. glabripennis) in the United States

Mitochondrial DNA sequence data can potentially yield insights into the history of ALB introductions. We present preliminary data that bear on the questions of origins and species recognition. Sequence data from mitochondrial COI support a clade for A. glabripennis from Jersey City, NJ, four sites within the quarantine zone of New York City (Flushing, Greenpoint, Maspeth and Sunnyside) and one specimen from Gansu, China. Another clade was formed from specimens originating in Chicago, IL, Austria, Long Island City, NY, and one specimen from Anhui, China. Specimens from Inner Mongolia and Korea were found in a different clade. These results support the accepted theory that Mainland China is the source of American populations and that separate introduction events occurred in New York City and Chicago. Adding sequence data from additional specimens is an ongoing process. Data support separate clades for the A. nobilis form of A. glabripennis and the A. malasiaca form of A. chinensis.. More informative morphological characters, or a second independent molecular data set, are needed to resurrect these species from synonymy.
