Mitochondrial DNA sequence divergence in Hessian fly

Domain III of the mitochondrial 12S rDNA and a region of the COII gene were examined in twenty populations and seven Purdue laboratory biotypes of Hessian fly. Nucleotide sequence data and PCR-RFLP revealed three haplotypes. A phylogenetic tree constructed using the neighbour-joining method supported three clades. Genetic distances ranged from 2.0% (haplotypes 1 and 2) to 6.4% (haplotype 3 and 1, 2). Most populations contained only haplotype 1 or the haplotype was present at high frequency (88%). Analysis of additional populations will reveal historical and phylogeographic patterns for Hessian fly in the United States, Canada and the Old World. Such knowledge will allow testing of the hypothesis that one haplotype was introduced into the New World from southern Europe, while the other two haplotypes were introduced from northern Europe. Results can have importance in revealing differences in selection by resistance genes in wheat during the dispersal and evolution of Hessian fly biotypes.
